Hello everyone! I will explain the streak plate technique. The streak plate technique is a microbiological method used to isolate pure bacterial colonies from a mixed culture. first, we take a sterile petri plate containing nutrient agar. then we sterilize an inoculating loop using a flame. After that we take a small amount of bacterial culture and gently streak it on the first quadrant of the agar plate. Next we sterilize the loop again and streak the second quadrant by dragging a few bacteria from the first quadrant. this process is repeated for the third and fourth quadrant. Each streak diluted the bacteria more and more.after incubation, separate colonies appear on the agar plate. Each colony grows from a single bacterium, which help to obtain a pure culture for further study. thank you!
